Description

Palnat 125 mg Capsule contains Palbociclib, a CDK4/6 inhibitor used in combination with hormonal therapies for the treatment of HR-positive, HER2-negative advanced or metastatic breast cancer. It slows cancer cell growth and improves progression-free survival in eligible patients. Palnat is an oral targeted therapy prescribed under oncologist supervision.

Indications and Usage

Palnat is indicated for the treatment of:

  • Hormone Receptor–positive (HR+), HER2–negative:

    • Advanced or metastatic breast cancer

    • In combination with:

      • Letrozole as initial endocrine-based therapy in postmenopausal women

      • Fulvestrant in patients with disease progression after hormone therapy

Must be used under strict oncologist supervision with routine blood monitoring.

How it Works

Palbociclib selectively inhibits CDK4 and CDK6, proteins responsible for promoting cell division in hormone-sensitive breast cancers. By blocking these, it halts the cancer cell cycle, slowing tumor growth and enhancing the effect of hormone therapies.

Dosage & Administration

  • Typical dose: 125 mg once daily for 21 days, followed by 7 days off (28-day cycle)

  • Should be taken with food at the same time every day

  • Swallow the capsule whole — do not open, crush, or chew

  • Continue endocrine therapy (letrozole or fulvestrant) as advised

  • Regular blood tests are required to monitor white cell counts and liver function

  • Dose may be adjusted based on side effects or blood test results

Side Effects

Common side effects:

  • Low white blood cell count (neutropenia)

  • Infections

  • Fatigue

  • Nausea

  • Diarrhea

  • Hair thinning

  • Mouth sores

  • Anemia

Serious side effects:

  • Febrile neutropenia (fever with low WBCs)

  • Severe infections

  • Lung inflammation (rare)

  • Liver enzyme elevation

Warning and Precaution

  • Not safe during pregnancy or breastfeeding

  • May suppress immunity — avoid contact with infections

  • Use contraception during and for 3 weeks after last dose (women), 14 weeks (men)

  • Monitor for signs of lung inflammation, bleeding, or fatigue

  • Avoid grapefruit and grapefruit juice

  • Inform your doctor about all ongoing medications
